One thing to know first
The Malay Heritage Centre has reopened after its revamp. Any guide still saying it is closed for redevelopment — including some of our own older copy — is out of date.
Bussorah Street, the pedestrian mall leading up to the mosque, is lined with cafes and shops selling perfumes, textiles and Middle Eastern food, while Haji Lane a block over is known for its mural-covered shopfronts and independent fashion boutiques. The area has no single opening hour since it's a neighbourhood of independent businesses, but most shops and cafes run from late morning into the evening, and the mosque itself asks visitors to dress modestly and to avoid entry during prayer times.
It borders Little India, about a 15-minute walk northeast.
Sultan Mosque closes to sightseers during the five daily prayer times, most disruptively around midday on Friday — if visiting the mosque itself rather than just photographing the exterior, check that day's prayer timings first.

The other good window. Rain tends to arrive early and clear, leaving usable afternoons. This is also when haze can occur, which is worth checking rather than assuming.
